That type of directory who is looking for your optimized title will ask for your url and fetch automatically, but generally in approx more than 90% of directories you can vary your title with your keywords.
like if your title is "SEO manual directory submission services - Get One way links".
You can use
Manual Submission Services
Manual Directory Submission services
Directory submission Services
Get One way links
and so on but keep in mind that you keywords include once with your title and generally try to avoid stop words with your title.

I would imagine that these days directories that allow keywords in your Title are probably highly devalued by Google. Just a thought.

It shouldn't be necessary to have the keywords in the title anyhow. The description should suffice.

Is there any harm or will cast a negetive impact on Google if i use all my keywords stuffed in the Title field, while Submitting to Directories.

Stuffing keywords anywhere is always a negative.
